According to Scott Gustin on X, Disney has confirmed the Red Car Trolley will close in 2025 to allow construction to begin on the Avengers Campus expansion, which necessitates the removal of the Red Car Trolley barn.

The above concept art shows the expanded Avengers Campus footprint, allowing for the upcoming fourth ride, Stark Flight Lab. The third attraction, Avengers: Infinity Defense, is also under construction. The addition of both rides is expected to double the size of Avengers Campus. No other closures to accommodate the expansion have been confirmed at this time

The expansion was announced at D23 earlier this month.

The Red Car Trolley opened in July 2012 with the full reimagining of Disneyland Resort’s second gate. It’s an iconic part of Buena Vista Street and the park in general. A closing date has not been announced.
